GenScript Upgrades OptimumGeneTM Gene Design System
GenScript's continuous improvement of its OptimumGene TM design platform has reached a new critical mass for gene synthesis and expression.
PISCATAWAY, NJ, November 05, 2008 /New Jersey News Articles/ -- CInternational life science contract research organization (CRO) GenScript Corporation continuously integrates new techniques and technology into its service platforms. GenScript's OptimumGeneTM gene design platform has reached new levels of efficiency and precision in gene synthesis and expression.
GenScript's OptimumGene TM system's primary purpose is to redesign genes of interest to improve protein expression in research settings. Principally, it can be very difficult to express a gene of interest in an organism not its own. For example, a laboratory may need to express a human gene in E. coli or cultured mouse cells. Many of GenScript's customers have found that altering the codon bias¡ªchanging the gene so that it codes for the exact same amino acids in a way better suited to the expression system¡ªcan make research quicker, easier, and less expensive. In addition, OptimumGeneTM mitigates the effects of problematic codons, and other problems.
The OptimumGene TM design platform employs a unique algorithm and proprietary codon usage table. The algorithm incorporates factors involved in gene transcription efficacy, translation efficiency, and refolding, such as GC content, CpG dinucleotide content, cryptic splicing sites, negative CpG islands, SD sequence, TATA box, terminal signal, codon usage bias, mRNA secondary structure, premature PolyA sites, internal chi sites and ribosomal binding sites, RNA instability motif, inhibition sites, stable free energy of mRNA, codon-context, the interactions of codon and anti-codons, and RNA secondary structures. Each of these can be adjusted to the customer's specific needs.
Additional restriction enzyme cutting sites can be added for later manipulation of the construct. In addition, the proteins expressed can be designed to incorporate epitope tag sequences (e.g., HIS or GST tag) or protease cutting sites for one-step purification. Libraries of variant forms of mutant genes can also be constructed for functional study and screening. Using this platform, viral genes can be expressed in mammalian cell lines, during which process the regulatory element in mammalian system is disrupted enabling the viral protein production to increase, and in some cases up to 28%. This may be of particular advantage to DNA vaccine development in that it promotes the immunogenicity of the target.
